Aiming at the Turtle Maker machine will do nothing, you must set your sights on the Turtles. Fire at the Turtles with the Shadow Blade, after the seventh Turtle is destroyed, the machine will self-destruct.

Unless you're using Game Genie codes, the only way to get this 1-UP is with Rush.
Use the Hard Knuckle to blast away the barriers to get the 1-UP.
Blast out the the bottom barrier with the Hard Knuckle, then the top barrier, then the middle barrier (so the Hard Knuckle can get recharged with the Large Weapon Pellet).
Shoot from here and jump when the bomb comes flying past.
Flying up here with Rush Jet is the easiest way to get to the ladder.
Slide in and get the Large Energy Pellets.
Shoot the Turtles with the Shadow Blade. Each Turtle with move faster than the last.
